بررسی اثرات طول دوره نوری بر فعالیت برخی آنزیم‌های گوارشی معده ای و روده ای در لارو و نوجوان قزل‌آلای رنگین‌کمان (Oncorhynchus mykiss)

هدف از این تحقیق بررسی دوره تکاملی آنزیم­های گوارشی و تاثیر دوره نوری بر فعالیت برخی آنزیم­های گوارشی معده­ای (پپسین)  و روده­ای(فسفاتاز­ قلیایی و -Nآمینوپپتیداز) در لارو و نوجوان قزل­آلای رنگین­کمان بود . تعداد 3600 لارو قزل­آلا در آغاز تغذیه فعال (18 روز پس از تفریخ) با میانگین وزن اولیه 009/0± 119 میلی­گرم در 4 تیمار دوره نوری و هر تیمار با سه تکرار (300 قطعه لارو در هر تکرار) قرار گرفتند و آزمایش طی 6 هفته انجام شد. 4 تیمار دوره نوری شامل 14 ساعت روشنایی و 10 ساعت تاریکی، 10 ساعت روشنایی و 14 ساعت تاریکی، 4 ساعت روشنایی و 20 ساعت تاریکی و 24 ساعت روشنایی طراحی گردید. نمونه­برداری از لاروها بصورت کاملاً تصادفی در روزهای0، 1، 3، 5، 8، 10، 15، 20، 25، 30، 39 و 44 روز بعد از آغاز تغذیه فعال انجام شد. طبق نتایج بدست­آمده، فعالیت آنزیم­های گوارشی در لارو و نوجوان ماهی قزل­آلا، تغییرات مشابهی با سن در همه تیمارهای نوری داشت. در انتهای آزمایش، فعالیت اختصاصی آنزیم پپسین و آمینوپپتیداز در تیمار نوری 24 ساعت روشنایی نسبت به تیمارهای 14 ساعت روشنایی، 10 ساعت روشنایی و 4 ساعت روشنایی بیشتر بود، اما اختلاف معنی­داری در فعالیت اختصاصی آنزیم­ فسفاتاز قلیایی بین دوره­های نوری مختلف مشاهده نشد((p>0.05. همچنین فعالیت آنزیم­های گوارشی لارو و نوجوان قزل­آلا، در دوره نوری طولانی­تر، بهبود می­یابد.

This study investigated the ontogeny of some digestive enzyme and explored the effects of photoperiod on stomach (pepsin) and intestine enzyme activity (alkaline phosphatase and N-amino peptidase) in rainbow trout (Oncorhynchus mykiss)larvae juvenile. About 3600 larval rainbow trout in first feeding (18 days after hatching) with an average initial weight 119±0.009 mg were distributed in plastic tanks in four treatments (300 larvae in each replicate). Four light treatments consisting of 14 to 10 h, 10 to 14 h, 4 to 20 h and 24 to 0 h (light: dark), were compared in triplicates for 6 weeks.. Finally, a random sampling from larvae was undertaken on days 1, 3, 5, 8, 10, 15, 20, 25, 30, 39 and 44 days after the onset of first feeding. According to the results, the digestive enzymes activity in fish larvae and juveniles had a similar change trend with age at all photoperiods. In addition, at the end of experiment, the specific activity of pepsin and N-amino peptidase were the highest in 24h Light treatment, but there was no significant difference in specific activities of alkaline phosphatase (p>0.05). The results demonstrated that growth and digestive enzymes activity of rainbow trout larvae and juvenile are improved by applying of longer photoperiod in rearing conditions.
